PSA self-test kits are often marketed as ‘convenient’, ‘simple’, ‘reliable’, and providing ‘reassurance’ to people who might be concerned about their risk of prostate cancer or their general prostate health. However, much of the marketing focuses on the issue of screening and testing for prostate cancer. Yet a PSA blood test is not a test for prostate cancer, but a test to help spot potential problems with the prostate. By itself, a PSA blood test cannot diagnose or rule out prostate cancer.

PSA is an important marker for prostate cancer screening. Combining serum PSA testing with a digital rectal examination can significantly increase the positive predictive value of prostate cancer.

PSA, as a specific marker in semen, can be used in forensic identification of semen stains. For example, at a crime scene or in a sexual assault case, detecting PSA in a semen stain can help confirm the presence of semen, providing clues for solving the case.

Most kits include equipment for you to collect a small sample of your own blood from your finger. These items include a finger-prick lancet (a small needle that can poke a hole in your skin), sterilising wipes, and plasters.
Some kits are designed for you to collect this blood sample in a small tube and send it off in a pre-paid envelope to the provider’s laboratory for testing. These results usually take between two and five working days.
The rapid test kits look like a COVID-19 test. You use a pipette to collect a few drops of blood from your finger-prick and drop a small amount onto the test cassette. The test gives you a result in five to 10 minutes.
Sample Collection&Storage
The PSA Rapid Test is intended for use with human whole blood, serum, or plasma specimens only.
Intake of Finasteride (5-reductaseinhibitor) will reduce the PSA concentration by max. 50%. This should be considered by the interpretation of the results.
Different factors could increase the PSA level in blood serum and should be avoided before collection of the blood sample and/or should be desisted from by the patient before sample taking.
Ride on a bicycle: 24 hours before taking of blood sample
Sexual activities (Ejaculation): 24-48 hours before taking of blood sample
Only clear, non-hemolyzed specimens are recommended for use with thistest. Serum or plasma should be separated assoon as possible to avoid hemolysis.
Perform testing immediately after specimen collection. Do not leave specimens at room temperature for prolonged periods. Serum and plasma specimens may be stored at 2-8°C for up to 3 days. For long term storage, specimens should be kept below -20°C. Whole blood collected by venipuncture should be stored at 2-8°C if the test is to be run within 2 days of collection. Do not freeze whole blood specimens. Whole blood collected by fingerstick should be tested immediately.
Containers containing anticoagulants such as EDTA, citrate, or heparin should be used for whole blood storage.
Bring specimens to room temperature prior to testing. Frozen specimens must be completely thawed and mixed well prior to testing. Avoid repeated freezing and thawing of specimens.
If specimens are to be shipped, pack them in compliance with all applicable regulations for transportation of etiological agents.
Icteric, lipemic, hemolysed, heat treated and contaminated specimens may cause erroneous results.
Safety of the PSA Test Kit
A BBC report has warned about the dangers of PSA self-test kits and how they can give men inaccurate results leading to anxiety and false reassurance. We recommend that if you’re worried about prostate cancer and want a PSA blood test, you should speak to your GP.
We haven’t tested the safety of any of the kits ourselves. However, some test kit providers state that they've been quality assured by the same accreditation schemes that would approve PSA testing via your GP. In the UK, this is usually the United Kingdom Accreditation Service (UKAS).
Medical laboratories that meet stringent requirements for quality and competence can be given ‘ISO 15189’ accreditation by UKAS, while ‘ISO 13485’ is the relevant standard for medical devices (such as PSA testing kits).
Some providers clearly state which accreditations they hold, or their laboratories hold, on their website. You might have to search around for this information, but it's sometimes listed in the FAQs section of the company website.
You could also check if a company has been accredited by UKAS, and whether the certificate that comes with the kit is valid, on the UKAS website. However, it's not always possible to find the company, or the laboratory they use, on this
Bring tests, specimens, buffer and/or controls to room temperature (15- 30°C) before use.
》Remove the test from its sealed pouch, and place it on a clean, level surface. Label the device with patient or control identification. For best results, the assay should be performed within one hour.
》Transfer 1 drops of serum/plasma to the specimen well (S) of the device with the provided disposable pipette, then add 1 drop of buffer, and start the timer.
OR
Transfer 2 drops of whole blood to the specimen well (S) of the device with the provided disposable pipette, then add 1 drop of buffer, and start the timer.
OR
Allow 2 hanging drops of fingerstick whole blood to fall into the center of the specimen well (S) of the test device, then add 1 drop of buffer, and start the timer.
Avoid trapping air bubbles in the specimen well (S), and do not add any solution to the result area.
As the test begins to work, color will migrate across the membrane.
》Wait for the colored band(s) to appear. The result should be read at 10 minutes. Do not interpret the result after 20 minutes.
