Rapid Test Uncut Sheet

1. Materials Required

- Nitrocellulose membrane: Core of detection zone

- Sample pad: Receives the sample

- Conjugate pad: Holds labeled conjugates (e.g., gold-labeled antibodies)

- Absorbent pad: Draws the liquid through

- Backing card (adhesive plastic film): Provides support

- Capture antibodies/antigens: For Test line (T-line)

- Control line antibodies (e.g., anti-IgG): For Control line (C-line)

- Conjugate (e.g., gold nanoparticles conjugated to antibodies): For signal detection

- Buffers, stabilizers, surfactants: For treatment and storage

2. Equipment Needed

- Dispenser/striping machine (for T-line/C-line)

- Conjugate spraying machine

- Laminating/assembly machine

- Cutter (optional if not cutting into strips)

- Incubator or drying oven (temperature-controlled)

- Desiccator or dry storage

3. Procedure

Step 1: Membrane Striping (Test & Control Lines)

- Stripe test and control lines on nitrocellulose membrane using micro-dispensing system.

- Dry at 37-45°C for 30-60 minutes.

Step 2: Conjugate Pad Preparation

- Impregnate with gold-labeled conjugate.

- Dry at 37°C for 2-4 hours.

Step 3: Pad Treatment

- Treat sample and absorbent pads with buffer solution.

- Dry thoroughly.

Step 4: Assembly

- Layer in order: Sample pad -> Conjugate pad -> NC membrane (with T/C lines) -> Absorbent pad.

- Ensure slight overlaps for capillary flow.

Step 5: Final Drying & Storage

- Dry entire uncut sheet at 37°C for 1-2 hours.

- Store in desiccator or sealed pouch with desiccant.

★Tips

- Conduct in cleanroom environment (Class 10,000 or better).

- Maintain low humidity (<30% RH) during drying and storage.

- Perform quality control on each batch (flow time, line intensity, etc.).
